Encapsulation efficiency is one of the most widely measured attributes for characterizing RNA-LNP formulations. It is also one of the most labor-intensive and variable to obtain.

Conventional approaches require detergent-mediated LNP lysis and around 8–10 preparation steps, creating multiple opportunities for operator error and inconsistent results.

About Aida López Espinar

Dr Aida López Espinar is a formulation scientist at Marama Labs focused on developing analytical methods for RNA-loaded nanoparticles. She holds a PhD in Pharmaceutics from University College Cork, where her research focused on the formulation and characterization of polymeric nanoparticles for extrahepatic mRNA delivery. Her research explores how Scatter-Free Absorption Spectroscopy can provide more accurate and reliable measurements from complex, light-scattering LNP formulations. She is the lead author of the study introducing CloudSpec’s integrating-sphere-based method for measuring RNA encapsulation efficiency without LNP lysis.

About Brett Rickeard

Brett Rickeard is a Field Application Scientist at Marama Labs focused on helping customers get the most out of their CloudSpec. He holds a MSc in Biochemistry from the University of Windsor, where his research focused on using scattering techniques to examine model lipid membranes. He’s authored several publications on lipid biophysics and conducted research at notable institutions around the world, including the Max Planck Institute of Colloids and Interfaces, Oak Ridge National Laboratory, NIST, and ANSTO. Brett spent several years in the LNP development group at Strand Therapeutics, before transitioning to field applications.
